Title: Optogenetic probing of sleep-wake circuits in the brain

Abstract:
The functions of sleep remains a mystery and the neural substrates involved are the subject of intense study. Successive sleep and wakefulness cycles rely on an appropriate balance between sleep-promoting nuclei of the brain, including inhibitory cells of the ventro-lateral preoptic area and anterior hypothalamus and, arousal-promoting nuclei from the brainstem and the posterior hypothalamus. Hence, the hypothalamus contains cell types able to regulate the occurrence and maintenance of sleep and wake events. Indeed, two distinct neuron populations restricted to the hypothalamus have been recently identified as critical modulators of the sleep-wake cycle: the Hypocretins- (Hcrt1,2, also known as OrexinsA,B)- and Melanin-Concentrating Hormone (MCH)-expressing neurons. We have previously shown that the Hcrt system is involved in promoting wakefulness. However, the role of MCH neurons on the sleep-wake cycle remains unclear. This presentation will summarize the application of in vitro and in vivo optogenetics to characterize the mechanisms implicated in the control of sleep state onset and maintenance.
