Are there changes in neuronal activity in non-stimulated regions of the
visual cortex? And do negative BOLD responses, which are pervasive in human
fMRI, reflect decreases in neuronal activity?
I will first demonstrate sustained decreases in fMRI signals and beyond the
stimulated regions in human retinotopic visual areas. The amplitude of this
negative BOLD response increases with increasing stimulus contrast. The
negative BOLD response is associated with decreases in cerebral blood flow
and with smaller decreases in oxygen consumption. Overall, findings in
humans indicate decreases in neuronal activity in non-stimulated regions of
early visual areas.
Next, based on experiments using simultaneous fMRI and electrical
recordings, I will characterize the neuronal correlates of the homologous
negative BOLD response in monkey V1. The negative BOLD response is
associated with decreases in neuronal activity, with comparable amplitudes
in the local-field potential and the multi-unit activity domains. The
cortical distances between the loci showing decreases in neuronal activity
and the activated regions indicate that the decreases in neuronal activity
cannot be solely mediated by the horizontal connections within V1. Finally,
using single-trial correlation, I will demonstrate a link between the
negative BOLD response and the corresponding decreases in neuronal activity.
 
The second part of my talk will focus on recent fMRI experiments in alert
monkeys, and on the methods that we have been developing to pursue them.
Functional brain imaging and physiology studies of the visual cortex often
utilize fixation on a spot as their baseline condition. It is not clear,
however, whether fixation per se causes changes in activity in early
retinotopic visual areas. Monkeys were trained to stay still during 20 s
long trials, during which they fixated on a spot. An increased BOLD signal
was observed in the foveal representation of V1 and V2 during trials in
which only the fixation spot was presented, with no additional stimulus. The
amplitude of this increased activity was largely invariant to different
contrasts (90%, 50%, and 10%) of the fixation spot relative to the
background. These findings indicate the involvement of attentional
mechanisms in early visual cortex during fixation.
